JPEG has less data than the raw, and compression removed some more, so in my view it is an inferior format is you are trying to measure things.
ISO doesn't change the light sensitivity of the sensor. It acts on an amplification level or the calibration of an ADC, or both. Different cameras have different techniques. And yes, the longer the exposure, the lighter the image.
For light intensity, get a physics book. But I would say that most of your assumptions are wrong, and a good scientific experiment starts with makre sure theere are no "assumptions", just hard facts.
